Olink Proteomics is a next-generation affinity-based protein quantification platform designed to enable large-scale biomarker discovery and validation in clinical research. Built on the proprietary Proximity Extension Assay (PEA) technology, the platform allows simultaneous measurement of hundreds to thousands of proteins from a minimal sample volume of just a few microliters. This combination of high multiplexing capacity and low sample input requirement makes Olink particularly well-suited for studies involving precious or limited biospecimens such as plasma, serum, cerebrospinal fluid, and urine.
Quantified protein levels are reported as NPX (Normalized Protein eXpression) values on a relative log2 scale, allowing straightforward comparison across samples and cohorts. The current flagship panel, Olink Explore HT, covers more than 5,000 human proteins in a single run, providing a near-proteome-wide view of the circulating proteome. Together, these features position Olink as a powerful tool for translational research, enabling the identification of disease-associated protein signatures and the development of clinically actionable biomarkers.

The evolution of protein analysis technologies, from classical immunoassays to modern high-throughput affinity-based methods
Traditional approaches such as mass spectrometry offer theoretically unlimited target breadth but remain constrained in throughput and sample volume efficiency, whereas multiplexed immunoassay platforms like Luminex and MesoScale Discovery enable simultaneous analysis of tens of proteins with moderate throughput. More recently introduced affinity-based platforms, particularly Olink PEA and SomaLogic, demonstrate superior performance across most categories, with Olink PEA achieving top-tier ratings in all six metrics including throughput, sensitivity, specificity, and the ability to analyze over 5,400 proteins from minimal sample volumes.
